These compounds inhibit cysteine proteases by forming covalent bonds with the active site thiol of cysteine proteases. In general, they are stable, and need the catalytic machinery of the cysteine proteases for activation. Of all the compounds developed, two Michael acceptor cysteine protease inhibitors, K-777 and Ruprintrivir (Fig. 1.2.6), have entered clinical trials.
